Standard 12 team, 6x6 category, head to head league: I got 4 players I'm choosing from to keep. Cano Fielder J Upton Granderson Cano is the only 1st round pick and the others are all 2nd rounders so if I took all of them they would be my 2nd, 3rd, and 4th round picks. Most of the league will have a keeper as a 1st rounder. Do I go for those 3 and hope to get lucky with a top pick in the 1st round with the chance to still get Cano or do I hold Cano and choose between the other 3. I'm leaning towards Cano, Fielder, and Upton.

You have it right. Granderson is the most likely to regress from last year so he is the easy cut.
